The study unveils for the first time how the key gene,\ndual-specificity\nphosphatase 22 (DUSP22),\nenhances T cell infiltration into tumours by \u2018trimming the stability tag\u2019 of\nthe immunosuppressive factor LGALS1. This discovery not only provides a novel\nperspective on tumour immune evasion but also proposes a promising combination\nstrategy to overcome immunotherapy resistance in \u2018cold\u2019 tumours,\nsuch as triple-negative breast cancer (TNBC). The findings have been\npublished in the <i>Journal for ImmunoTherapy of Cancer<\/i>, the official\njournal of the Society for Immunotherapy of Cancer (SITC).<\/p><p>In the tumour microenvironment, the scarcity of CD8\u207a T cells\u2014a primary\ntype of anti-cancer\nimmune cell\u2014is a fundamental reason for the poor efficacy of immunotherapies\nagainst many solid tumours. Transforming these immune-desert \u2018cold\u2019\ntumours into treatment-sensitive \u2018hot\u2019 tumours is a key\nchallenge in cancer immunotherapy today. To address\nthis, the team used\ngenome-wide screening techniques to identify\n39 candidate genes associated with T cell infiltration in a mouse\nmodel of breast cancer. Among these, DUSP22 emerged as the most prominent gene.\nExperiments confirmed that the\nlevel of DUSP22 in breast cancer tissues positively correlates\nwith CD8\u207a T cell infiltration and negatively correlates\nwith malignant tumour progression. Crucially, overexpressing DUSP22 in tumour\ncells significantly suppressed tumour growth and prolonged the survival\nof mice\nwith a\ncompetent immune system, an effect that was completely absent in\nimmunodeficient mice. This is akin to discovering a \u2018switch\u2019 within tumour cells that regulates\nimmune cell infiltration into solid tumours. The\nteam also\nelucidated the\nmechanism by which DUSP22 specifically removes phosphate groups from two\nkey sites (Ser8\/Thr58) on the immunosuppressive molecule LGALS1. This\ndephosphorylation destabilises\nLGALS1, leading to its rapid degradation. As LGALS1 typically induces T cell apoptosis\nand hinders their ability to cross blood vessels and enter\ntumours,\nits depletion relieves immunosuppression, allowing T cells to infiltrate tumours\nunimpeded and kill cancer cells efficiently.<\/p><p>Based on this mechanism, the team further explored its clinical\ntranslational potential. They\nfound that LGALS1 inhibitors (such as OTX008) and neutralising\nantibodies not only inhibit tumour growth independently, but\nalso demonstrate a\nsignificant synergistic effect when combined with the widely used\nanti-PD-1 immunotherapy. This combination therapy\nsuccessfully transformed\nimmunotherapy-resistant \u2018cold\u2019\ntumours into responsive \u2018hot\u2019\ntumours, offering a new avenue to overcome immunotherapy resistance. This\nresearch expands the function of DUSP22 beyond traditional signal transduction\nregulation, establishing its crucial role in the \u2018crosstalk\u2019 between tumours\nand the immune system. It also highlights the critical role of LGALS1\nphosphorylation in tumour immune evasion.<\/p><p>The corresponding authors of this study are Prof\nDeng\nand Prof Miao. The first author is Wang Lijian, a PhD\nstudent\nin UM FHS. The research team also includes scholars from UM,\nthe University of Hong Kong\u2013Shenzhen Hospital, the Macau\nUniversity of Science and Technology, and the Third Affiliated Hospital\nof Zhengzhou University.
