Standardised cryopreservation technique is\nused to freeze tumour tissue specimens obtained during early surgical\nprocedures. In cases where tumour samples are unavailable during recurrence or\nmetastasis, the frozen tissues can be revitalised for organoid culture, providing\nsupport for potential personalised treatment options for cancer patients or\nfurther cancer research. The research has been published in the international\njournal <i>Bioactive Materials<\/i>.<\/p>\n\n<p>A biobank involves the\ncollection and storage of substantial quantities of biological specimens, along\nwith associated personal data such as medical records, family history,\nlifestyle information, and genetic data. In recent years, there has been a\nnotable increase in the number of large-scale biobanks being established,\nincluding those for tumour tissues and other samples. These biobanks typically\nstore specimens under cryogenic conditions using ultra-low temperature freezers\nor liquid nitrogen. Unfortunately, these storage methods often kill the cells. The\nspecimens can then only be used to extract proteins, DNA, or RNA, and cannot be\nused to cultivate live cells. This significantly limits their overall value and\npotential applications.<\/p>\n\n<p>To address this\nchallenge, the UM research team proposed the novel concept of establishing a biobank\nof living tumour tissues. They developed a standardised technique in which the\ntissues are gradually cooled and transferred to liquid nitrogen for long-term\npreservation. This technique redefines tissue biobanking, as it enables the\ncultivation of patient-derived tumour organoids (PDOs) from cryopreserved\ntissues, while supporting air-liquid interface (ALI) cultures and preserving\nthe original tumour immune microenvironment. This technique successfully\nmaintains tissue viability, and holds great promise for advancing personalised\nprecision cancer treatment and research.<\/p>\n\n<p>In the study, the research\nteam tested 42 pairs of frozen human tumour specimens. Tissue samples from four\ndifferent cancer types\u2014breast cancer, colorectal cancer, lung cancer, and\nkidney cancer\u2014were used to establish PDOs. The results showed that organoids\ncould be successfully cultivated in 40 out of 42 cases, achieving a success\nrate of 95.2%. Notably, the tumour samples could still be used to establish\norganoid models even after nearly four years of cryopreservation. The study also\ndemonstrated the applicability of this technique for effectively cryopreserving\nsmall specimens, including biopsy tissues.<\/p>\n\n<p>This living tissue\ncryopreservation technique is simple and easy to operate. It enables various\ntumour specimens to be frozen long term while maintaining the viability of the cells\nwithin the tumour tissues. Organoids derived from these tissues can be routinely\nfrozen and passaged, enabling high-throughput screening of drug sensitivity and\nidentification of potentially effective drugs to combat drug-resistant tumours.\nMoreover, cryopreserved tumour tissues retain the original tumour\nmicroenvironment, making them suitable for ALI cultivation and evaluation of\nanti-tumour drug responses, including immune checkpoint inhibitors. The study\nexpands the concept of biobanking and lays the foundation for establishing\nliving tissue biobanks, providing substantial support for precision cancer\ntreatment and translational medical research.<\/p>\n\n<p>Prof Deng is the\ncorresponding author of the study, and Chen Ping, research assistant professor in\nFHS, is the first author. FHS postdoctoral researchers Zhou Jingbo and Feng Yangyang\nand FHS doctoral student Chu Xiangpeng also made significant contributions to\nthe study.
