{"id":413914,"date":"2025-06-11T15:07:00","date_gmt":"2025-06-11T07:07:00","guid":{"rendered":"https:\/\/www.um.edu.mo\/news-and-press-releases\/campus-news\/detail\/61317-2\/"},"modified":"2025-06-18T12:54:45","modified_gmt":"2025-06-18T04:54:45","slug":"61317","status":"publish","type":"post","link":"https:\/\/www.um.edu.mo\/pt-pt\/news-and-press-releases\/campus-news\/detail\/61317\/","title":{"rendered":"Equipa de investiga\u00e7\u00e3o da UM encontra liga\u00e7\u00e3o entre a elimina\u00e7\u00e3o de res\u00edduos cerebrais e a psicose precoce"},"content":{"rendered":"\n                        \n                        <p>A research team led by Yuan Zhen, head of the Centre for Cognitive and\nBrain Sciences (CCBS) and professor in the Faculty of Health Sciences (FHS) at\nthe University of Macau (UM), has for the first time identified dysfunction in\nthe glymphatic system, the brain\u2019s waste clearance system, among patients with early\npsychosis. The team also offered a novel neuroimaging biomarker for the early\ndiagnosis and intervention of psychiatric disorders. The research has been published\nin <i>Molecular Psychiatry<\/i>,\na leading international journal in neuroscience and psychiatry.<\/p>\n\n<p>The glymphatic system plays a vital role in clearing metabolic waste and\nmaintaining neural health. Previous studies have shown that dysfunction of the glymphatic\nsystem is closely linked to neurodegenerative diseases such as Alzheimer\u2019s and\nParkinson\u2019s. However, systematic studies and clear evidence on its involvement\nin psychiatric disorders, particularly in early psychosis, are lacking.<\/p>\n\n<p>In this study, the research team analysed multimodal\nMRI neuroimaging data from 137 participants aged 16 to 35. These participants\nincluded healthy controls, individuals with non-affective psychosis, and individuals\nwith affective psychosis. The team measured the coupling between the cortical\nblood-oxygen-level-dependent (BOLD) signal and the ventricular cerebrospinal\nfluid (CSF) flow, which served as a biomarker for the brain\u2019s waste clearance\nefficiency.<\/p>\n\n<p>The results showed that BOLD-CSF coupling was\nsignificantly weakened and delayed in patients with early psychosis, suggesting\nglymphatic dysfunction. Patients with affective psychosis exhibited the most\npronounced abnormalities, indicating that their glymphatic systems may be\nimpaired at an early stage of the disease (Fig. 1). Further analysis revealed\nthat reduced BOLD-CSF coupling correlates with declines in\nfluid intelligence, concentration, working memory, and language skills, as well\nas higher anxiety levels. The reduced BOLD-CSF coupling may also be closely related to sleep disorders and chronic\ninflammation, which are commonly associated with psychosis, reflecting the\ncondition\u2019s extensive impact on the glymphatic system.<\/p>\n\n<p>The study also found that the dysfunction was most\npronounced in high-order cognitive networks (e.g. default mode and\nfrontoparietal networks), while lower-order sensory and motor regions (e.g.\nvisual and somatomotor networks) were less affected. This suggests a \u2018top-down\u2019\npattern of impairment in early psychosis (Fig. 2). This pattern indicates that\npsychosis may first damage the brain regions responsible for complex cognitive\nprocessing before affecting the regions responsible for sensory processing.<\/p>\n\n<p>In addition, the proposed BOLD-CSF coupling index demonstrated high\nstability and reproducibility across multiple independent datasets (Fig. 3), showing\nstrong reliability across different platforms and populations. It will be a\npromising neuroimaging biomarker for psychiatric disorders, which can enhance\nearly detection, disease progression monitoring, and therapeutic response\nassessment, thus improving the objective and quantitative diagnosis and\nintervention of psychiatric disorders.<\/p>\n\n<p>The corresponding authors of the study are Prof Yuan and Zhao Zhiying, research\nassistant professor in CCBS. The first author is doctoral graduate Hua Lin.\nDoctoral graduate Zeng Xinglin and master\u2019s graduate Zhang Kaixi also\ncontributed to the study.
