A research team\nled by Wang Chunming, professor in the Institute of Chinese Medical Sciences (ICMS)\nat the University of Macau (UM), has developed a \u2018glue\u2019 derived from\nglucomannan that is designed to repair damaged intervertebral disc tissue,\noffering a promising new approach to alleviating IDD. The research has been\npublished in the international journal <i>Nature\nCommunications<\/i>.<\/p>\n\n<p>As people grow older, degeneration of the intervertebral\ndiscs can lead to common symptoms such as chronic lower back pain and reduced mobility\nin the lower limbs, significantly diminishing quality of life. However, current\nclinical treatments either only provide temporary pain relief or carry a risk\nof recurrence following surgery. Through the detailed analysis of clinical\nspecimens and databases, the research team made a groundbreaking discovery: the\nprogression of IDD is linked to the loss of a protein called MFG-E8, which is\ncrucial for maintaining the health of nucleus pulposus cells. Further\ninvestigation revealed that this protein depletion is caused by an increase in glycosidases\nin the nucleus pulposus tissue of the disc. These enzymes act like scissors, cleaving\nthe glycosaminoglycans (GAGs) that normally protect MFG-E8. Building on this finding,\nthe team synthesised a glucomannan octanoate (GMOC), a compound with a structure\nsimilar to human GAGs, which can shield MFG-E8 from degradation and resist\ndestruction by glycosidases.<\/p>\n\n<p>The researchers developed GMOC into a \u2018polysaccharide\nglue\u2019 and tested it in two animal models of IDD (rats and rabbits). The results\nshowed that injections of the polysaccharide glue effectively slowed disc degeneration\nand alleviated pain symptoms. The research has been patented in China and\ninternationally through UM, and efforts are underway to translate it into\nclinical applications through industry-academia collaboration. If successfully\napplied in clinical settings, GMOC could potentially reduce the need for\ninvasive surgery. This study is one of the team\u2019s many achievements in their decade-long\nresearch on the use of polysaccharides, oligosaccharides, and\nglycosaminoglycans for tissue repair and regeneration.<\/p>\n\n<p>The corresponding author of the study is Prof Wang, and\nthe first three authors are PhD student Liu Yu, and master\u2019s students Chao Tzuwei\nand Li Yuwei, all from ICMS. The research also received substantial support\nfrom Dong Lei, professor in the School of Life Sciences at Nanjing University,\nand the research team of Geng Dechun, professor at the First Affiliated\nHospital of Soochow University.
