The study unveiled a two-component chemical defence system\nin <i>Panax<\/i> <i>notoginseng<\/i> mediated by chloroplast-localised <i>\u03b2<\/i>-glucosidase and 20(<i>S<\/i>)-protopanaxadiol\nginsenosides, which will contribute to a deeper understanding of how <i>Panax<\/i> species defend against pathogens\nand provide valuable insights for the development of sustainable botanical\npesticides. The research results have been published in the internationally\nrenowned journal <i>Nature Communications<\/i>.<\/p><p>Plants produce\nsecondary metabolites as a response to various environmental threats, enabling\nthem to adapt and play a crucial role in chemical defence. The research\noriginated from unexpected findings during a quality control study of <i>Panax notoginseng<\/i> leaves. After meticulous\ninvestigations, the research team unveiled a two-component chemical defence\nsystem in <i>Panax notoginseng<\/i> leaves. The\nsystem comprises a chloroplast-localised <i>\u03b2<\/i>-glucosidase,\nPnGH1, and its substrates, 20(<i>S<\/i>)-protopanaxadiol\nginsenosides. The <i>\u03b2<\/i>-glucosidase and\nits substrates are spatially separated in cells under physiological conditions.\nGinsenoside hydrolysis is activated only when exoenzymes produced by pathogenic\nfungi disrupt the plant\u2019s chloroplast. The activation of PnGH1-mediated\nhydrolysis produces a series of less-polar ginsenosides through selective\nhydrolysis of outer glucose at the C-3 site. These newly formed ginsenosides\nexhibit a broader spectrum and more potent antifungal activity. The researchers\nalso found a similar two-component<i> <\/i>chemical\ndefence system in other <i>Panax<\/i> species,\nsuch as ginseng (<i>P. ginseng<\/i>), and American ginseng (<i>P. quinquefolium<\/i>). These findings shed light on the development of botanical\npesticides and are hoped to reduce reliance on chemical pesticides in the\ncultivation of <i>Panax<\/i> species, thus\ncontributing to the advancement of research in traditional Chinese medicine and\nthe sustainable development of the industry.<\/p><p>The\ncorresponding authors of the study are Wan Jianbo; Shi Shepo, researcher from\nthe School of Chinese Materia Medica at Beijing University of Chinese Medicine;\nand He Xiahong, professor at Southwest Forestry University. The co-first\nauthors are Ma Lijuan, postdoctoral researcher at UM, and Liu Xiao, associate\nresearcher at Beijing University of Chinese Medicine.
