{"id":289657,"date":"2023-01-18T15:27:00","date_gmt":"2023-01-18T07:27:00","guid":{"rendered":"https:\/\/www.um.edu.mo\/news-and-press-releases\/press-release\/detail\/55015-2\/"},"modified":"2023-01-18T17:43:40","modified_gmt":"2023-01-18T09:43:40","slug":"55015","status":"publish","type":"post","link":"https:\/\/www.um.edu.mo\/pt-pt\/news-and-press-releases\/press-release\/detail\/55015\/","title":{"rendered":"Joint study of UM and two universities of Chinese medicine helps alleviate sepsis-induced injury"},"content":{"rendered":"\n                        \n                        <p>A research team jointly\nled by Elaine Leung Lai Han, professor in the Faculty of Health Sciences (FHS)\nof the University of Macau (UM); Xie Ying, professor at the Second Affiliated\nHospital of the Guangzhou University of Chinese Medicine; and Chen Jianxin, professor\nat the Beijing University of Chinese Medicine has discovered that the modern\nherbal formula BaWeiBaiDuSan (BWBDS) can help alleviate\nsepsis-induced liver injury (SILI). The study has received considerable attention in the\nfield of pharmacology and the research results have been published in the\ninternationally-renowned journal <i>Acta Pharmaceutica Sinica B<\/i>.<\/p><p><a name=\"OLE_LINK354\"><\/a><a name=\"OLE_LINK355\">Sepsis is widely perceived as a\nlife-threatening organ dysfunction caused by a dysregulated host response to\ninfection. Antibiotics are an essential component of initial therapy in sepsis.\nHowever, the inappropriate use of antibiotics can lead to multidrug-resistant\ninfections and death. It is necessary to discover novel targeted or combined therapeutic\nstrategies. The gut-liver axis refers to the bidirectional relationship of microbiota,\nmetabolic, and immune crosstalks between the gut and liver, connected in a\nbidirectional fashion by the portal vein and biliary tree. SILI is related to\nadverse clinical outcomes and gut microbiota plays a crucial role in SILI. The formula BWBDS is based on\ncomponents extracted from eight medicinal herbs, namely <i>Panax ginseng C. A.\nMeyer<\/i>,<i> Lilium brownii F. E.\nBrown ex Miellez var. viridulum Baker<\/i>,<i> Polygonatum sibiricum Delar. ex\nRedoute<\/i>,<i> Lonicera japonica Thunb.<\/i>,<i> Hippophae rhamnoides Linn<\/i>,<i>\nAmygdalus Communis Vas<\/i>,<i> Platycodon grandiflorus (Jacq.) A. DC.<\/i>,<i> <\/i>and\n<i>Cortex Phellodendri<\/i>. Previous studies by the team have showed that traditional\nChinese medicine (TCM) formulas with <\/a>medicine food homology can improve intestinal microecology and enhance host immune function. Therefore,\nin the study, the team investigated whether the BWBDS treatment could reverse\nSILI by modulating the gut microbiota. The study shows that BWBDS and <i>L. johnsonii <\/i>may act as prebiotics and\nprobiotics for the prevention and treatment of sepsis.<\/p><p>Although the clinical\nefficacy of BWBDS is significant, the pharmacological mechanism of BWBDS\nremains unclear. The team performed qualitative and quantitative analysis of\nBWBDS components with high performance liquid chromatography-tandem mass\nspectrometry (LC\/MS\/MS). Network pharmacology was applied to predict possible\nmechanisms of BWBDS. Cecal ligation and\npuncture (CLP) was employed to induce sepsis and related injuries caused by\npost-operative infection. Faecal microbiota transplantation (FMT) and 16S\nPacBio single-molecule real-time (SMRT) sequencing were used to validate the\nroles of gut microbiota. Metabolomics analysis was performed to characterise\nthe metabolic profile. Clodronate liposome and anti-interleukin-10 receptor\nmouse antibody (anti-IL-10R mAb) were used to explore the potential mechanism\nof bacteria attenuating SILI.<\/p><p>In the study, BWBDS\nselectively promoted the growth of <i>Lactobacillus<\/i> in CLP-treated mice.\nResults of the FMT treatment show that gut bacteria are correlated with sepsis\nand are required for BWBDS antisepsis effects. The treatment with <i>L.\njohnsonii <\/i>significantly prevented mice from sepsis and SILI and improved\nthe survival of mice, which is associated with enhanced gut\nintegrity, as well as reduced intestinal and systemic inflammation. In\nparticular, the research results suggest that the reduction of CLP-induced mortality\nand alleviation of liver injury by <i>L. johnsonii<\/i> depend on the promotion\nof the proportions of IL-10<sup>+<\/sup>M2 macrophages. Moreover, the\nstudy shows that heat inactivation <i>L. johnsonii <\/i>(HI-<i>L. johnsonii<\/i>)\ntreatment can promote the anti-inflammatory\nactivity of macrophages and alleviate SILI.<\/p><p>The team\u2019s findings provide a novel therapeutic\napproach for SILI. In the\nforeseeable future, gut microbiota status, such as species diversity, will be\nused as a biomarker to predict the therapeutic effects of BWBDS in sepsis patients. In addition, BWBDS and\nthe gut bacterium <i>L. johnsonii<\/i> can be translated to clinical use as novel\nprebiotics and probiotics for the\ntreatment of sepsis patients. The study has revealed\nthe great potential of TCM in treating sepsis by regulating gut microbiota.<\/p><p>Prof Leung, Prof Xie, and Prof Chen are the co-corresponding\nauthors of the study. PhD students Fan Xiaoqing and Mai\nChutian at the Macau University of Science and Technology and PhD student Zuo Ling\nat the\nBeijing\nUniversity of Chinese Medicine are the co-first authors.
