{"id":277354,"date":"2022-09-27T14:48:00","date_gmt":"2022-09-27T06:48:00","guid":{"rendered":"https:\/\/www.um.edu.mo\/news-and-press-releases\/press-release\/detail\/54228-2\/"},"modified":"2022-09-28T11:43:52","modified_gmt":"2022-09-28T03:43:52","slug":"54228","status":"publish","type":"post","link":"https:\/\/www.um.edu.mo\/pt-pt\/news-and-press-releases\/press-release\/detail\/54228\/","title":{"rendered":"UM research team successfully develops near-infrared light-emitting carbon nanodots for biomedical imaging"},"content":{"rendered":"\n                        \n                        <p>A research team led by\nQu Songnan, professor in the Institute of Applied Physics and Materials\nEngineering, University of Macau (UM), has proposed a strategy to prepare\nnear-infrared (NIR) carbon nanodots by fusing large conjugated molecules. Using\nthis strategy, the team has successfully prepared carbon nanodots with the main NIR luminescence\ncenter in aqueous solution\nfor the first time in\nthe world. The preparation cost of the carbon nanodots is very low, less than\none percent of the cost of commercial NIR dye ICG. Moreover, the carbon\nnanodots are safe and non-toxic with good biocompatibility and have been\nsuccessfully applied to NIR <i>in vivo<\/i>\nimaging and ear angiography in mouse models. This study provides new insights\nfor the development of safe and low-cost <i>in\nvivo<\/i> NIR fluorescence imaging reagents. The research results have been\npublished in the internationally renowned academic journal <i>Advanced Science<\/i>.<\/p><p>Compared with visible\nlight (400-700nm), NIR light (700-1700nm) has longer wavelengths and can penetrate\ndeeper into biological tissue, making it more suitable for biological imaging.\nCarbon nanodots are a new class of nano-luminescent materials, which have\nattracted much attention due to their low production cost, facile synthesis,\nand low biological toxicity. However, the emission wavelengths of the reported\ncarbon nanodots are mainly in the visible light region, and regulating the\nemission bandgap of carbon nanodots has always been a research hotspot in this\nfield. Through literature research, the research team found that the bandgap of\ncarbon dots synthesised from small conjugated molecules such as\nphenylenediamine can be adjusted to the red band, but it is difficult to reach\nthe NIR band. Theoretical calculations show that the optical bandgap of\nconjugated carbon dots with a particle size of about 2 nm can reach the NIR\nband, and most of the carbon dots reported so far exceed this size, indicating\nthat the effective conjugation degree of these carbon dots is insufficient.\nThus, it is difficult to achieve a narrow NIR emission bandgap.<\/p><p>Combining theoretical\ncalculation results, the group proposed a strategy of fusing large conjugated\nmolecules to prepare NIR luminescent carbon nanodots. The team members\ncondensed perylenetetracarboxylic dianhydride molecules containing five benzene\nrings with urea molecules through solvothermal condensation. The main\nabsorption peak and main emission peak of the prepared carbon nanodots in the aqueous\nsolution are both in the NIR band, and the luminescent efficiency in the NIR\nband is as high as 18.8 per cent, which is the highest efficiency of carbon\nnanodots reported so far. In the <i>in\nvivo<\/i> imaging experiment, the bright NIR fluorescence signal emitted from\nthe mouse intestine after gavage for one hour showed a high signal-to-noise\nratio (S\/N) of 18, proving that the carbon nanodots have the ability of deep\ntissue NIR imaging. Furthermore, two-photon NIR FL angiography of the mouse ear\nwas performed with two-photon fluorescence emission under 1300 nm femtosecond\nlaser excitation. The signal of carbon nanodots (red signal) reached its\nhighest intensity 15 seconds after injection, and blood vessels in the mouse\near were clearly visible.<\/p><p>Prof Qu is the corresponding author of this study.\nThe first authors are Liu Yupeng and Wang Gang, two PhD students in the IAPME,\nand Lei Haipeng, a postdoctoral fellow in the Faculty of Health Sciences.
