{"id":273890,"date":"2022-08-17T17:13:00","date_gmt":"2022-08-17T09:13:00","guid":{"rendered":"https:\/\/www.um.edu.mo\/news-and-press-releases\/campus-news\/detail\/53921-2\/"},"modified":"2022-08-17T17:13:00","modified_gmt":"2022-08-17T09:13:00","slug":"53921","status":"publish","type":"post","link":"https:\/\/www.um.edu.mo\/pt-pt\/news-and-press-releases\/campus-news\/detail\/53921\/","title":{"rendered":"UM study discovers promising therapeutic target in prostate cancer"},"content":{"rendered":"\n                        \n                        <p>A research\nteam led by Edwin Cheung, professor in the Faculty of Health Sciences (FHS), University\nof Macau (UM), has reported the discovery of a novel protein factor responsible for promoting prostate\ncancer growth. The result of a study conducted by the team suggests that this\nprotein factor is a promising therapeutic target in prostate cancer as inhibiting\nits expression will significantly affect the occurrence and progression of the\ndisease. The research result was recently published in the internationally\nrenowned journal <i>EMBO Reports<\/i>.<\/p><p>Prostate cancer is the most common type\nof cancer found in men and is also the second-leading cause of cancer death among\nmales. The androgen receptor (AR), a hormone-regulated master transcriptional\nregulator, is responsible for the initiation and progression of prostate cancer.\nAR transcriptional activity is meticulously controlled by the physical and\nfunctional interactions with transcriptional co-factors. Changes in the expression\nor activity of any of these co-factors in prostate cancer will significantly affect\nthe occurrence and progression of the disease. Thus, identifying and understanding\nthe role of AR co-factors in prostate cancer is key to developing novel diagnostics\nand therapeutic strategies to combat this ailment.<\/p><p>In this study, the research team used\na proteomics technique known as Rapid immunoprecipitation mass spectrometry of\nendogenous proteins (RIME) to systematically identify all proteins\ninteracting with AR in prostate cancer cells. They found that Tripartite motif-containing-motif TRIM33 (TRIM33) is a critical\ncomponent of the AR transcriptional regulatory network, acting as a positive regulator\nof AR-mediated transcription. They also demonstrated through a series of detailed\nmechanistic experiments that AR recruits TRIM33 to specific genomic locations and\nstabilises AR protein levels by protecting AR from Skp2-mediated ubiquitination\nand proteasomal degradation. These events ultimately lead to enhanced AR\nbinding to chromatin and the transcriptional activation of downstream target genes\nthat control DNA metabolism, cell cycle, mitosis, and prostate tumor-related\nsignaling pathways. The results indicate that TRIM33 plays a tumour-promoting role\nin prostate cancer.<\/p><p>Prof Cheung\u2019s team analysed the\nclinical data from prostate cancer patients. First, they found a significant\nincrease in TRIM33 mRNA and protein levels in human prostate tumour tissue compared\nto normal prostate tissues. The researchers then defined an AR target gene directly\nco-regulated by TRIM33, and found that this gene set was highly expressed in\nprostate tumours and that patients with high expression of this gene signature\nare at a higher risk of disease recurrence. Therefore, the inhibition of TRIM33\nexpression makes prostate cancer cells more sensitive to AR antagonists. Together,\nthese results suggest that TRIM33 is a promising therapeutic target in prostate\ncancer.<\/p><p>Prof Cheung is\nthe corresponding author of the study. His PhD students, namely Chen Mi and\nShreyas Lingadahalli, as well as former research assistant Nitin Narwade, are\nco-first authors of the study. FHS Associate Professor Terence Poon Chuen\nWai and his team also made important contributions to this study.
