{"id":260647,"date":"2022-02-07T15:30:00","date_gmt":"2022-02-07T07:30:00","guid":{"rendered":"https:\/\/www.um.edu.mo\/news-and-press-releases\/press-release\/detail\/53196-2\/"},"modified":"2022-03-16T12:13:00","modified_gmt":"2022-03-16T04:13:00","slug":"53196","status":"publish","type":"post","link":"https:\/\/www.um.edu.mo\/pt-pt\/news-and-press-releases\/press-release\/detail\/53196\/","title":{"rendered":"UM desenvolve uma nova rede met\u00e1lica-fen\u00f3lica para melhorar a efic\u00e1cia da imunoterapia"},"content":{"rendered":"\n                        \n                        <p>A research team led by Assistant Professor Dai Yunlu in the\nUniversity of Macau (UM) Faculty of Health Sciences (FHS) has developed a novel\nmetal-phenolic network to potentiate the activation of stimulator of interferon\ngenes (STING) pathway within tumour microenvironment for advanced radiotherapy.\nThe novel nanomedicine is expected to improve the efficacy of immunotherapy. The\nresearch findings have been published in <i>Advanced Materials<\/i>.<\/p>\n\n<p>In recent years, new developments in immunotherapy have\nrevolutionised cancer treatment. However, the \u2018cold\u2019 tumour microenvironment\ncaused by low tumour-infiltrating lymphocytes results in a low response rate of\ncancer immunotherapy. Recent studies have shown that the activation of STING\npathway can activate innate immunity and turn a \u2018cold\u2019 T cell-infiltrated\ntumour microenvironment into a \u2018hot\u2019 one, which can help to improve the\nefficacy of immunotherapy for oncology patients. Traditional radiotherapy\ngenerates cytosolic dsDNA to evoke STING pathway activation. Additionally, MnCl<sub>2<\/sub>\nalone can act as an activator of the STING pathway. The research team\nengineered a novel metal-phenolic network via the coordination interactions\nbetween phenolic ligands and metal ions, potentiating STING pathway activation\nwithin tumour microenvironment for advanced radiotherapy.<\/p>\n\n<p>In this study, the amphiphilic polymer PEG-polyphenol was\nintroduced to coordinate with lanthanide-doped radiosensitizer (DSNPs) and Mn<sup>2+<\/sup>\nto form stabilised metal-phenolic networks DSPM. After cell internalisation, DSPM\ndisassembled owing to the cleavage of imine linkers in PEG-polyphenol,\nimparting the release of DSNPs and Mn<sup>2+<\/sup>. Upon X-ray irradiation,\nDSNPs sensitised cancer cells and promoted cytosolic dsDNA release. The\nreleased Mn<sup>2+<\/sup> facilitated the recognition of cytosolic dsDNA by\ncGAS, triggering the activation of the STING pathway and DC maturation.\nSubsequently, tumour-specific antigen was presented to T cells to evoke robust\nantitumor immunity. The 4T1 tumour-bearing mice model showed that DSPM combined\nwith X-ray irradiation elevated the recruitment of tumour-infiltrating\nlymphocytes and the secretion of proinflammatory cytokines, promoting the\nsuccessful transformation of the nonimmunogenic \u2018cold\u2019 microenvironment into an\nimmunogenic \u2018hot\u2019 tumour microenvironment.<\/p>\n\n<p>Prof Dai is the corresponding author of the study.
