{"id":220878,"date":"2021-01-12T16:27:00","date_gmt":"2021-01-12T08:27:00","guid":{"rendered":"https:\/\/www.um.edu.mo\/news-and-press-releases\/press-release\/2021\/01\/51257-2\/"},"modified":"2021-03-09T15:07:58","modified_gmt":"2021-03-09T07:07:58","slug":"51257","status":"publish","type":"post","link":"https:\/\/www.um.edu.mo\/pt-pt\/news-and-press-releases\/press-release\/detail\/51257\/","title":{"rendered":"UM desenvolve novo nanoprocessador para o tratamento de cancro"},"content":{"rendered":"\n                        \n                        <p>Assistant Professor Dai Yunlu\u2019s research team\nin the University of Macau\u2019s (UM) Faculty of Health Sciences (FHS) has designed\nand synthesised an oxygen-enriched metal-phenolic X-ray nanoprocessor to\nenhance the therapeutic effects of anti-tumour radiotherapy. The nanoprocessor can\nimprove radiodynamic therapy and enhance anti-tumour immune response by focusing\nthe ionised energy generated by radiotherapy in the tumour microenvironment while\ngenerating ROS to induce cancer cell damage. The study has been published in\nthe top international academic journal <i>Advanced Science<\/i>.<\/p><p>Radiotherapy is a traditional cancer\ntreatment that has been proven to be effective in all types of cancer treatment.\nHowever, the toxicity of high doses of radiation to normal tissues and the\ninhibition of toxic substances during radiotherapy limit the effectiveness of\nradiotherapy. To overcome this limitation, the research team developed an\noxygen-enriched X-ray nanoprocessor: Hb@Hf-Ce6 nanoparticles, which can be used\nin combination with PD-1 antibody to improve the therapeutic effects of\nradiotherapy and radiodynamic therapy and enhance anti-tumour immune response.<\/p><p>Hb@Hf-Ce6 nanoparticles are prepared based on\nthe structure of metal polyphenol coordination, which integrates all functional\nmolecules into nanoparticles to achieve multifunctional anti-tumour effects.\nThe polyphenol structure modified by chlorin e6 (Ce6) is coordinated with\nhigh-Z radiotherapy sensitisers (hafnium, Hf), and the oxygen carrier\nhemoglobin (Hb) is chimeric in the coordination structure to overcome radiation\ntolerance associated with hypoxia. Specifically, under X-ray radiation, the\nradiation-induced physical process stimulated by Hf can activate the\nphotosensitiser Ce6, thereby generating high levels of ROS to induce cancer\ncell damage.<\/p><p>The results of an <i>in vitro<\/i> breast cancer 4T1 cell experiment showed that Hb@Hf-Ce6\nnanoparticles enhanced tumour cell death induced by radiotherapy and radiodynamic\ntherapy. In addition, the introduction of immune checkpoint inhibitor enhanced immune\nresponse induced by radiotherapy and radiodynamic therapy in 4T1 orthotopic and\nbilateral tumour model, thus achieving efficient and long-term inhibition of\ntumour growth and metastasis. In conclusion, this combined therapeutic strategy\ninduces a comprehensive anti-tumour immune response targeting cancer\neradication and metastasis inhibition. This study proposes a multifunctional\nmetal polyphenol nanoplatform, which can be combined with immunotherapy to achieve\nefficient X-ray-mediated radiotherapy-radiodynamic therapy, to provide new therapeutic\noptions for cancer treatment.<\/p><p>\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n\n<\/p><p>Prof Dai is the corresponding author of the\nstudy.
