Faraz Mohammadali Shaikh,\na doctoral student from the University of Macau (UM) Faculty of Science and\nTechnology (FST), has developed compounds that destabilise Ebola during his\nrecent internship at the Wellcome Trust Centre for Human Genetics (WTCHG),\nUniversity of Oxford, United Kingdom. \u2018It\u2019s motivating to be part of the\nproject and chart a path to end this epidemic,\u2019 he says.<\/p><p><br\/><\/p><p><b>Work with Oxford Research Group<\/b><\/p><p><b>\n<br\/>\n<\/b>During his six-month internship, Shaikh participated in a drug design\nproject against the Ebola virus. \u00ad is project was headed by Prof David Stuart,\ndirector of Oxford\u2019s Division of Structural Biology. The world-class research\ngroup at the division is focused on using innovative experimental and\ncomputational techniques to solve the complex problems of structure-function\nrelationship of viruses of different sizes, as well as the puzzles of\nvirus-receptor interactions and virus assembly.<br\/>\n<br\/>\nShaikh\u2019s job was to work on the computational methods for designing drugs\nagainst Ebola. He used computer simulations and fragment-based molecular\ndocking to explore viral activity \u2013 in particular how the virus enters the host\ncell \u2013 at the atomic level, and to investigate ways to effectively inhibit\nvirus attachment and membrane fusion. The goal of this research project is to\nidentify chemical entities as potential candidates for developing drugs against\nEbola.<\/p><p><b><br\/>\nStunning Learning Outcome<br\/>\n<br\/>\n<\/b>Ebola is a highly virulent pathogen which can cause a severe haemorrhagic\nfever with a high death rate. In four months, the research group scanned 9\nmillion compounds through computational analysis in order to obtain compounds\nthat can bind to Ebola and destabilise Ebola in thermos-stability assay. \u2018I\ndidn\u2019t have much time at Oxford and the clock was ticking, so I proceeded\neverything in one shot. We\u2019ve got some compounds that work. And I\u2019m so proud\nand thrilled that four compounds from my list were purchased by Oxford from the\nsame private company. The cost of each compound differs. All compounds cost\naround 1,000 USD in total,\u2019 says Shaikh.<br\/>\n<br\/>\nWhat Shaikh achieved at Oxford far exceeded his expectations. In the beginning,\nShaikh just wanted to see how the Oxford research group executed their work.\nThen he performed his task very well and contributed to a new discovery. \u2018We\nhave also got the Crystallography structure of compound bound to Ebola\nglycoprotein,\u2019 he says. Decreasing the stability of GP the inhibitor could\nprevent the fusion of the virus with endosomal membranes. We are currently\nfinalising the results and will publish a paper in some good impact journals\nfor this collaboration between UM and Oxford. We will keep working on Ebola\nuntil we get something.\u2019<\/p><p><br\/>\n<b>Way to Oxford<br\/>\n<br\/>\n<\/b>As to how to get accepted by Oxford, Shaikh hints that it is not about\ncompetition. He attended an interview with Prof Stuarts and Prof Yvonne Jones.\n\u2018It lasted about 30 minutes, with questions related to my previous experience\nand computational drug design skills, and that is it. Fortunately, things went\nsmoothly,\u2019 Shaikh says.<\/p><p><br\/><\/p><p>\n<b><\/b><\/p><p><b>Solid Research Experience at UM<br\/>\n<br\/>\n<\/b>Shaikh attributes the internship to his PhD supervisor Siu Weng In, an\nassistant professor from the Faculty of Science and Technology. \u2018It is Prof Siu who bridged me there. She has given\nme a lot of constructive advice that has beneted both my life and my studies.\nShe has been guiding me no matter where I am,\u2019 says Shaikh. Also, Prof Siu\u2019s\nstrong background in computational biology often benefits him. After obtaining her PhD degree from Saarland\nUniversity in Germany, Prof Siu joined UM and has since led a research group in\nbioinformatics. \u2018\u00ad e way she works and instructs inspires me. Each new day\nworking with the group takes me one step closer to my goal \u2013 improvise\nsomething that matters.<br\/>\n<br\/>\nShaikh admits that it is quite risky for professors to send their students\nabroad for more than half a year. \u2018\u00ad at might delay the research project she\nhas planned with me. And yet, Prof Siu is optimistic, supportive and contented\nwith whatever I work on in the lab there.\u2019 Probably, thanks to such an\ninspirational and friendly ambience at UM, Shaikh has been able to maintain his\nenthusiasm and exceed his own expectations. \u2018Prof Siu often encourages me to\ntake more challenges and her expertise in algorithm and molecular docking\noffers me a very solid training in the first year at UM. And that has had a\ngreat impact on my research visit to Oxford. Without my professor, it would not\nhave happened,\u2019 he says.<\/p><p>\n\n\n\n\n\n\n\n\n\n\n\n<\/p><p><br\/>\n<b>Grow with UM<br\/>\n<br\/>\n<\/b>Shaikh was at a crossroads in his life when he considered studying for a\ndoctoral degree. Choices were many. In the end, Shaikh decided to study at UM\nas he sees UM as a growing university. He prefers to be with the team where he\nneeds to do a lot of setup tasks. \u2018I don\u2019t want to work for a well-established\ngroup. I want to progress with the team. I\u2019ll apply what I have learned from\nOxford to my work here.\u2019<br\/>\n<br\/>\n\u2018UM has made a comprehensive analysis of the capability of each student. We are\nalways given the opportunity to execute whatever projects we have in mind.\nAlso, we\u2019d gain a lot of support and resources from UM as long as the project\nruns well. \u00ad at actually makes UM a fast-growing university,\u2019 says Shaikh. \u2018In\njust one and a half years, I\u2019ve seen a lot of progress achieved here.
